Transformation into Thermococcus

  • Grow overnight cultures of TK
  • add plasmid, buffer
  • heat shock
  • spread onto -trp plates
  • incubate in canister with anaerobic pack for 3-5 days
  • Pick cultures into epe tubes filled with .8X asw
  • spot each culture to a discreet portion of a lovastatin plate
  • Incubate for 3-5 days
  • Remove 20 cultures and put each of them into a of complete ASW media, containing
    1. ASW liquid media
    2. Lovastatin
    3. Agmatine
    4. Sulfur
    5. KOD vitamins
  • Place in incubator until grown (supposed to be 12-16 hr)
