<p>Obesity is an increasingly prevalent human condition in the world. Despite major progress in the understanding of the molecular mechanisms leading to obesity, no safe and effective treatment has yet been found. Herein we aim to utilize the RNA interference(RNAi) technology to silence the anti-apoptotic gene BCL2 specifically in white adipose tissue, leading to the ablation of white fat. We use an endogenous nanoparticle exosome on account of its low toxicity and immune response to deliver the nucleotide drug. The exosomal surface will be modified with an adipose tissue-specific circular peptide tPep(CKGGRAKDC) by a glycine-linker to the lysosomal-associated membrane protein 2b(Lamp2b) which is a protein found abundant on the surface of exosomes. The selective delivery of tPep-exosome-siRNA to white adipose will theoretically promote the adipose cell death, ultimately representing a potential approach for the treatment of obesity.</p>
