Biologists tend to use One Factor At a Time (OFAT) approaches when trying to determine important factors for a process or system, or when attempting optimisation. OFAT can have some large disadvantages, including an inability to identify interactions between factors. This could lead to two or more factors being identified as non-important, however if both/all were removed this may have an effect. An example of this is if two factors are providing the same function, if one is removed the other will still provide that function, however if both were removed then the functionality would disappear.

A solution to this would be a full factorial approach, where every combination of factors is tested (Figure 1), however this becomes unfeasible when dealing with a large number of factors due to the amounts of experiments required. Design of Experiments (DoE) is a common engineering approach used to analyse factors in a process/system in order to perform optimisation. DoE is capable of multifactorial analysis and uses mathematics to determine the fewest combinations (experiments) required to obtain statistically relevant results. JMP [REF] is a software package which can determine experimental designs using DoE and analyse the results. Here, DoE has been used to optimise the supplement solution for Cell-Free Protein Synthesis (CFPS) systems.
