    <p>Now that we have plenty of Enteromorpha to deal with, we focus on the cellulose and hemicellulose which don’t have much added value in themselves thus wasn’t made good use of. And what we have done is fermentating them into ethyl alcohol.

    Co-culture system is one biological system which has a great potential, especially in the aspect of its cooperative fermentation. Fiber corpuscle, one of the scaffold protein, is an existing efficient system in the nature. But in yeast, its expression may make the host metabolic burden too heavy. So we think of using E. coli to replace fibre corpuscle, which plays a similar effect to increase the efficiency of the enzyme.

            <p style="font-size:20px"><strong>After hearing the event, <i>S.cerevisiae</i> and <i>E.coli</i>decide to come and defeat <i>Enteromorpha</i>.<br/><i>E.coli</i> uses cellulase and hemicellulase as his sword to smash <i>Enteromorpha</i>.<br/>The fragment of <i>Enteromorpha</i> becomes cellodextrin and xylose.<br/>At the same time, <i>S.cerevisiae</i> collects the fragment and make it into glucose and then, ethanol.<br/>Finally, mixing with resveratrol, <i>S.cerevisiae</i> transforms the monster into Enteromorpha wine. </strong>  </p>

            <p style="font-size:20px"><strong><i>Enteromorpha</i> disappeared!<br/>Now every summer, people enjoy themselves on the beautiful beach in Qingdao!</strong></p>
After pretreatment, we successfully transformed the cellulose and hemicellulose into cellobiose and xylose that are easier to use.

The cellobiose is then transformed into ethyl alcohol through the intrinsic pathway of saccharomyces cerevisiae .

As for the xylose,we transform it into xylulose and ultimately the ethyl alcohol.

Thus we created two kinds of functional yeast which can live on special carbon source ---when we feed them with cellobiose and xylose, they will produce ethyl alcohol in return. What’s more, we can adjust the ratio between two yeasts to accomplish the best output corresponding to the original composition of Enteromorpha. Amazing!

      <p>In addition to Enteromorpha contains numerous cellulose and hemicellulose, there are a large number of antioxidant polysaccharide substances. These polysaccharide substances can be used as materials for health care products. At the same time, we use the yeast to express resveratrol, an antioxidant substances either, to improve our antioxidant health care products.

    <p>When we compare the expression of eukaryotic and prokaryotic genes,eukaryotic gene expression vectors are larger because of single cistron,lengthy promoters and terminators.Therefore,we try to build a short and strong expression combination(mini-system).It contains a small enough promoter and terminator.In addition, we tested to ensure optimal combination of promoter and terminator.

A Bottle Of Algae "Wine"
Producing ethanol from algae


This year OUC-iGEM focuses on the efficient use of cellulose. We are concerned about the utilization of Enteromorpha as a hot local issue. Every year a large quantity of Enteromorpha are salvaged ashore. The utilization, however, remainds still in the initial stage . Compared to the traditional strategy of using terrestrial plant cellulose, the ingredient of lignin in large seaweed is rather low, which can lead to a better degradation.So it will be a great idea to produce new energy from Enteromorpha.
Our project can be devided into 3 parts:


Adhesion platform

E. coli and yeast is to adhere to each other and form a multi-cell unit, expanding the biological function of engineered cells.

Fermentation

We aim to make use of cellobiose and xylose produced from algae residue and turn them into ethanol---our Enteromorpha wine! Resveratrol is added to our wine to make it better for health.

Mini-GRE

We synthesize mini promoters in yeast, which have very concise sequences. In this way, we look forward to making our own contribution to the standardization of yeast!
