This summer, we participated in the CCiC (Conference of China IGEMer Community) which was held by a non-profit committee in China. It brought together many new and old IGEMers, professionals, scholars, students etc and provides a precious platform for academic exchanges in terms of synthetic biology. With sufficient preparation and confidence, we set foot in Fujian Agriculture and Forestry University where the forth CCIC was situated. With excitement, the knowledge feast started.

At the beginning of the meeting, Dr Ling and Dr Zhang Haoqian from blue SPAR made the opening speech. Then they discussed over the concern that how our students of biology major adapt themselves in such a rapidly developing society in which knowledge changes everyday and find their own future. During the three-day-meeting with a tight timetable, teams from around the national gave a speech with PPT in showing of their IGEM projects. After that, it came to the defense part where particular professional scholars would judge and evaluate our projects. In presentation, we began to get nervous at first, but it faded quickly. In the defense of King l.c., our project was confirmed, which also ensure our confidence in success. At the same time, each team brought posters and promote their projects to visitors. We were inspired by the superb ideas of other teams which widened our scope of knowledge and horizons.

Delighted and honored, professor Kenneth A.O from IGEM HP and professor King L.C. was invited to participate in the meeting. They told apart the classification of HP and answered the confusion about bio-safety. Researchers like Lou Chunbo and Fu Xiongfei also shared many valuable experiences with us. These let us stand on the shoulders of giants and see the wonderful world of science.

On 29th July, SCUT-CHINA_B team invited us to the South China University of Technology for a meeting. Both teams introduced their project to each other and questions and opinions had been raised during discussions. After meeting, SCUT-CHINA_B team kindly gave us a tour around the school campus. This was our pleasure to meeting them and have a chance to communicate with undergraduates team.

</br></br>Keeping track of all the things we have done, summarizing the things we have learnt, reflecting on our action and sharing our knowledge with the public make our project much more meaningful. We established an IGEM column within the official account of Shenzhen BGI-college on the Wechat platform in March. Since then, we have documented many of our stories.</br></br>

More than half of the members of our team have participated in writing articles and publishing them after being revised by our instructor and advisors. Now we have 8 articles published, covering the status quo of lung cancer molecular diagnostics, iGEMers meet-ups, synthetic biology, iGEM competition judging and discussion on our project’s future prospective. </br></br>

To promote and gain support for our project, we initiated crowdfunding at different platforms.

At the beginning, we started small. We participated in the High School Crowdfunding Festival on the WeChat (Chinese Social Network) Platform and raised more than ¥9,360+additional rewards for popularity ¥316=¥9,676(approximately $1,500) within three days under the project name “A new lung cancer diagnosis method.” Then we stated our mission and how we would accomplish them: ”We want to create a bio-sensor  used with test paper to provide a harmless, efficient and precise lung cancer gene detection method, increasing the survival rate of the patients.” </br></br>

</br></br>Then we went to the largest crowdfunding website in China, zhongchou.com, and more detailedly documented our project.  The goal has been set at ¥30,000 (approximately )

When presenting our project, we started with the introduction of synthetic biology first. Next, we moved to discuss EML4-ALK and the improved paired-dCas9 system. Then we touched a little bit on the design of the test paper. Finally, we introduced the team members and clearly listed how we would use the money we get. More than 110 people have supported us, many leaving encouraging short comments for us. </br></br>

Describing our project in a vivid and understandable way to those unfamiliar with synthetic biology is not an easy task. When trying to do this, we realized we did not understand the whole project as well as we thought. So here comes the question, by how much can high school students get involved in this competition themselves, especially the design part? How much can they really absorb and how far can they reach? How much help should they receive from the advisors? Do the time they spend on learning really match what they receive? These are all good questions to be considered, and we asked them on Zhihu (China’s quora). </br></br>

</br></br><h3>ICG-12</h3></br></br>

We luckily got a chance to attend pop science carnival in ICG-12 (The 12th International Conference on Genomics) held in Shenzhen, China. One of our team members gave a presentation on “omics for all” section which introduced what is iGEM to the public. The presentation also discussed what was our project, what we had done and what we were doing at that time. In the end, we advised teenagers to try to know what their dream career actually are and be prepared for the future.

<h3>Education.</h3>

Many of us wouldn't be able to enjoy iGEM if not for the enthusiastic lectures on synbio and iGEM given by former iGEM-ers. To thank them, we collaborated with the Shenzhen Foreign Language School team to give small science lectures to enrich STEM education in primary school and middle school.

To our surprise, the kids in primary school knew more about genes and gene detection than we thought! It turned out that they had done researches beforehand. We told them the story of the discovery of DNA’s structure and gave them a small test afterwards. Almost all of the children got all the questions correct, and we gave them postcards and badges as presents. One interesting moment was after we mentioned Rosalind Franklin’s study and how she was unable to justify her place in the discovery because of her identity as a female scientist, all the students chosen by their teams to participate in the bio quiz round were girls!

</BR></BR>In the middle school, we gave a talk on cancer’s cause on the genetic level (oncogenes) and how gene detection could be used to aid prognosis. We managed to let them know the CRISPR/Cas9 system in terms of where it was from, how would the Cas9/dCas9 proteins work and where this system could be applied. (and then gave our project as an example.)

</br> Team members in Shenzhen were actively preparing for another presentation of iGEM and our project after Giant Jamboree, hoping to establish a new team in the Shenzhen Senior High School, one of the largest high school in the city. We had discussed the content with the biology teacher and hoped for a good result at GJ can help us attract new iGEMers~

<h3>Filming video on biosafety</h3>

Considering there is no existing video introducing biosafety in laboratory for educating new iGEMers and the public, we decided to film our own THIS IS BIOSAFETY video. A five minutes video shows a story between the boss and two novices, reveals more than15 common mistakes that can be made in laboratory and demonstrates the correct approach in laboratory. This video has been uploaded on Chinese and foreign social media where every people can watch it!

Integrated human practice


We investigate 10 Chinese companies who provide ctDNA detection or cancer screening services. The investigation on current ctDNA detection services among 10 Chinese companies gives us a vision on the diversity of detection services and market share in China.
Conclusion:



Each company has its unique method to carry out service. It suggests there is no technology monopoly in this industry. Therefore our product can compete with others in technology aspect as we have our own method to detect lung cancer.

The time required to complete the service is between 2~8 days, except for EZlifeBio. It is seen to me that our product have a clear advantage in the length of time required because the detecting process can finish within few hours and the result can be read straight away. Similarly, EZlifeBio also sell detector and kit to customers and customers can run the detecting process. However, our target audiences are different. Their detectors are targeting hospitals or laboratories but our product is targeting the end user, patients, in this industry.

The outcome from each service is slightly vary but all of them provide an assistance in cancer diagnosis or cure. It is distinct to our product as we aiming to provide a straightforward result for customer to understand. However, our product shows its limitation in the result compare to other outcomes from those services. It needs to be developed to generate more completed result in order to compete with other services.

Communication with BGI product research department

Before we start to design our product, we consulted a staff who works in BGI product research department. She gave us a lot of advice on how to design a product and what should be the main concerns during the development of a product. Her suggestions inspired us to create a simple screening paper for lung cancer detection.

After we introduced and explained our project to him, he gave his opinions from a professional point of view. First, he questioned about the efficiency of using cell-free system to complete a series of reactions and the extremely low concentration of ctDNA in patient’s blood. Then, he also concerned about applying our design onto a screening chip instead of other product forms such as a detection kit. After that, we exchanged our opinions on those questions with Dr. Fang Zhengyu. It is greatl to have others opinion on our project. Dr. Fang’s suggestions helped us improve the design of our product and we decided to carry out further experiments to teat the efficiency of the screening paper.

Abstract



According to our investigation above, there are already all kinds of ctDNA detecting services existed in the market. Therefore, it is hard to compete with those developed products. Our well designed paper chip has its own unique selling points which can open up a new small market. It is aiming to provide a fast and efficient method to detect ctDNA in individuals, leading to an early diagnosis of lung cancer or a monitor on cancer recurrence.

Market analysis



1. lung cancer in China

1.Trends in Incidence Rates (Age-Standardized to the Segi Standard Population) for Selected Cancers for Males:

China, 2000 to 2011.



Among the 10 most common cancers considered in the temporal trend analyses for men, lung cancer always has the highest incidence rate in China between 2000 and 2011, contributed almost 0.05% each year.



2.Trends in Incidence Rates (Age-Standardized to the Segi Standard Population) for Selected Cancers for Females: China, 2000 to 2011.

For women, even though lung cancer has a lower incidence rate than that of men, it still has the second


highest incidence rate in China.

3.Trends in Mortality Rates (Age-Standardized to the Segi Standard Population) for Selected Cancers for Males: China, 2000 to 2011.



A similar trend can also be found in the mortality rate of lung cancer compares to the incidence rate trend for males. The lung cancer mortality rate for female experienced a slightly decrease to just below 0.01% in 2011.

From the graphs above, lung cancer can be seen as one of the biggest threat to Chinese people’s health, with both high incidence and mortality rate. Only minor differences can be found between the incidence and mortality rate for both males and females indicate that lung cancer is still a life-taking disease with a low chance to be cured in individuals. One of the reason for this is lung cancer always can only be detected and diagnosed when it is already developed into stage 3 or 4. At this stage, cancer has spread into rest of the body which causes the difficulty to cure. Our product can provide a simple, low cost, and efficient way to carry out early lung cancer detection. It is also easy to use which individuals can detect their ctDNA for lung cancer by themselves before going to hospital for a more detailed check.

Both tables show a huge proportion of lung cancer patients is found in the rural areas in China. Those areas may have their limitation in public health care and medical services. Our product can provide a cheaper and easier way to detect ctDNA which can helps the early diagnosis of lung cancer.

According to statistic data from ministry of public health, Chinese health market account for 373 million person-time in 2014. It had increased 62.17% compare to 230 million person-time in 2009. However, Chinese still has a relatively low percentage,27.27%, of people having health check each year as the population accounts for 1.368 billion people in China.

Chinese professional health check market capacity is 1.6 billion yuan in 2015. It is predicted that the CAGR of the capacity of health check market will be 25% between 2016 and 2020. Health check market capacity is projected to reach 5.2 billion yuan in 2020. As people are more concern about their health, the health check market has a bright prospect.

It connects with pharmacy and medical equipment companies and individual customers who comes to health check. At the moment, the service level and the market occupancy of different health check organization are become more and more distinct. It can be found earlier than other symptoms of lung cancer, which gives an advantage in early cancer detection. -The result is clear and easy to understand. Weakness: -At the moment, our product has it limitation on detecting a variety of cancer. This product only aim for EML4-ALK mutation in lung cancer. Only 4–6% of lung cancer involve this fusion gene. External attributes of the organization Opportunities: -According to the discussion on health check market above, China has a huge health check market with sustained growth annually. Our product is easy to used and efficient which is suitable to be used in health check. Unlike other lung cancer detection method, our screening chip provide detection services for a massive number of patients in a period of time. Therefore it has its advantage to be in the health check market. -People are more aware to have regular health check to prevent diseases. More people accept the concept of “early discover, early diagnosis, and early cure”. Therefore an additional option for lung cancer detection during health check match the demand from patients. Threats: -Public may distrust our product due to lack of understanding of our product. Therefore education is necessary before promoting the product.

Conclusion



Overall, our product has it potential outcompete other lung cancer services and fill the demand in the market. The strengths of the product give it advantages to access the health check market but the weakness should be improved for our product to be used widely. We have to seize the opportunity in health check market but before that, wide promotion and education are necessary to prevent misunderstanding from public.
