We have registered a great team and had agreat summer for processing our project. For now we are now looking forward to the GiantJamboree.

We have filled out all the forms and pages thatare required carefully. Also, we prepared our poster and presentation to bettershow you guys about our project.

The data of the validated parts we made anduse widely in our project is as follows:(parts的链接)

This year we have construct a lot of parts,here we show the three best new parts we did: The first one is ‘Rpa-Lasconverter’ (http://parts.igem.org/Part:BBa_K2315046), which can convert Rpamolecules into Las molecules; the second one is ‘LasR-pLas-GFP HSL induciblefluorescent actuator’ (http://parts.igem.org/Part:BBa_K2315034), which can beinduced by several AHLs and test their crosstalk; the last one is ‘Las moleculegenerator’, which can produce Las 3OC12-HSL without inducer. We have successfullycharacterization these three new parts and submitted the data to the main page.Finally we have submitted them to the iGEM Parts Registry on time.

We have collaborated with other iGEM teamsin meaningful ways and made contributions to projects of other teams.

* We worked as a leader and collaboratedwith other 10 iGEM teams to take videos about biosafety.

* We provided USTC a kind of reagent – CdSnanoparticles for testing whether CdS nanoparticles can increase thecathode-current as expected. Eventually we received their feedback of their data.

* We helped UCAS characterize theirnitrogen promoter to make their data more solid.

* We visited Fudan and finally gave them arealistic model for their project.

* We collect parts from other teams to helpus for constructing our devices library.

* We have attended many conferences and havemeetings with many iGEM teams. We have participated in CCiC in FAFU, and one ofour team member, Mingzhe Chen, has be invited to give a short introductionabout quorum sensing system.

The great amount of work we have done forHuman Practices has given rise to the increasing interests of Synthetic Biologyof at least hundreds of adults from different fields. Firstly we have donepublic education: In this part, we brought synthetic biology and MagicBlock tothe public. We visited high schools to introduce synthetic biology to thestudents and to guide them to design bio-product using MagicBlock. We also usedSocial media to publicize synthetic biology and our project. Secondly, weleaded biosafety videos for 10 iGEM teams: Biosafety is one of the mostimportant issues everyone should be aware of before starting any biologyrelated experiments. However, the biosafety education in China is far behindthe fast development of modern biological research. In some places, studentsare not trained properly because of the outdated education material or thelanguage barrier that prevent them from reading biosafety guidelines written inEnglish.

Our integratedhuman practice constitutively guides our project in various ways:

From one hand,inspired by the creativity and enthusiasm of non-biologists through interviewsand questionnaires, we have found that many people are very enthusiastic andimaginative about bio-designs, yet have little knowledge in synthetic biologyand little lab experience.

During theinvention of MagicBlock, we maintain contact with the public, to testMagicBlock as well as receiving suggestions. we held two workshops, guidingparticipants to use MagicBlock to design their own bio-products. We received variousthought-provoking designs using MagicBlock elements, such as water pollutionalarm, synthetic music, and bacteria painting panel .et al, all originated fromnon-biologists.

During thispractice, we received valuable suggestions to improve MagicBlock .

From the otherhand, Professional Insights we’ve received by professors, provide guidelinesfor our follow-up works including human practice, hardware and software design,and modeling.

we simplify the actual biology process intobasic model that only remains input molecule, promotor, transcription gene,mRNA, goal protein and output molecule from both dynamic perspective andresponding ability. In developed model, we consider different conditionsincluding the population growth, diffusion of signal and decay of signalmolecules in cells. which will have influence on our block. Finally, wecompletely construct the model of our block, which will instruct our experimentresults and using of our system. Moreover, this model does some basicresearches on population and new measurement methods.

In the result section, we have proved that eachof our parts (hardware, software, wet lab, model and human practice) works verywell as we expected! In another word, it can be safely concluded that our MagicBlock system works very well!
