Cancer is one of the most deadly diseases in the world. Cancer accounts for more than 8.8 million deaths in 2015. Lung cancer, one of the top five cancers, took away 1.69 million lives in 2015. Although medicine and cancer treatments have been highly developed overtime, it is still difficult to diagnose and cure lung cancer in its early stage, particularly for non-small cell lung cancer (NSCLC).

Our main task is to construct a test paper that contains a powerful biosensor capable of detecting any DNA sequence. The biosensor, which is composed by dcas9 and T7 polymerase coupled system, can act like a smart automatic switch. When the test paper detects a particular DNA sequence, it then releases a visible signal.

Cancer is one of the most deadly diseases in the world. Cancer accounts for more than 8.8 million deaths in 2015. Lung cancer, one of the top five cancers, took away 1.69 million lives in 2015. Although medicine and cancer treatments have been highly developed overtime, it is still difficult to diagnose and cure lung cancer in its early stage, particularly for non-small cell lung cancer (NSCLC).

Our main task is to construct a test paper that contains a powerful biosensor capable of detecting any DNA sequence. The biosensor, which is composed by dcas9 and T7 polymerase coupled system, can act like a smart automatic switch. When the test paper detects a particular DNA sequence, it then releases a visible signal.

For diagnosing NSCLC cancer, we will use our biosensor to detect the fusion gene EML4-ALK, which is thought to have huge connections with lung cancer. The formation of this protein has to do with the alteration that occurred in the chromosomes. The alteration means one small piece of chromosome mysteriously going to the tail of another chromosome. If we are able to use our dcas9 and T7 polymerase coupled system to find EML4-ALK effectively and identify it at an early stage, then we will be creating a faster and safer cancer diagnostic process and immensely help people in pain with NSCLC cancer or even other types of cancers.

We have another envision as well: enabling our biosensor to spot potentially dangerous experimental  cells and initiate cell self-killing process when the cells escape its original habitat. This could build a much safer experimental environment. Our detection system is promising because through designing different target sequences for the biosensor it can be applied to detection of many other types of cancers or genetic diseases.

<p>There were experiments done in support of the relationship between the protein EML4-ALK with small cell lung cancer (SCLC) or non-small cell lung cancer (NSLC). Many experiments used real-time PCR and screening to detect the existence of the protein in the samples. An experiment discovered that the isoforms of the protein EML4-ALK has direct connections with NSCLC, which was found in two patients (1). There were also other evidences tested from reverse transcription PCR screening that endorsed the idea of strong associations between EML4-ALK with SCLC or NSLC (2). One of the existing remedies for this kind of lung cancer is Alectinib. It is an oral drug that block ALK activities and applied as one of the most recent targeted therapies that aim at the EML4-ALK fusion gene.

<p>The BGIC-Union is founded under BGI (The Beijing Genomics Institute). Our team consists of 12 Chinese students from different cities in China, UK and USA. <p>From June to August, all the members will come together and complete most of the experiments, which is one of the most crucial parts of our project. At the same time, we will also begin to work on human practices, which is to “go beyond the lab”, and extend our project into a social context. We will explore our project’s relation with ethical, social, economic and bio-safety issues and bring it to the next level. Lectures and events, which are based on our project, will be delivered in schools or in public. We will raise public awareness on lung cancer, synthetic biology and precision medical treatment.

In November, BGIC-Union will fly across half of the world and take part in the Giant Jamboree in MIT to present our project. In the long term, we hope our project can go further, spark new thoughts in genetic engineering and inspire more young people to do more research!

DESCRIPTION

Cancer is one of the most deadly diseases in the world. Cancer accounts for more than 8.8 million deaths in 2015. Lung cancer, one of the top five cancers, took away 1.69 million lives in 2015. Although medicine and cancer treatments have been highly developed overtime, it is still difficult to diagnose and cure lung cancer in its early stage, particularly for non-small cell lung cancer (NSCLC).

Our main task is to construct a test paper that contains a powerful biosensor capable of detecting any DNA sequence. The biosensor, which is composed by dcas9 and T7 polymerase coupled system, can act like a smart automatic switch. When the test paper detects a particular DNA sequence, it then releases a visible signal. For diagnosing NSCLC cancer, we will use our biosensor to detect the fusion gene EML4-ALK, which is thought to have huge connections with lung cancer. The formation of this protein has to do with the alteration that occurred in the chromosomes. The alteration means one small piece of chromosome mysteriously going to the tail of another chromosome. If we are able to use our dcas9 and T7 polymerase coupled system to find EML4-ALK effectively and identify it at an early stage, then we will be creating a faster and safer cancer diagnostic process and immensely help people in pain with NSCLC cancer or even other types of cancers.
