Since our project is based on optogenetic tools to manipulate the neuron activity, we submit several basic parts which is essential to the function. The part collection provides other teams with materials to neuron manipulation and visualisation, if other teams require these parts, they can refer to iGEM databases.
In order to express functional channelrhodopsins and indicator in specific nueron-AWA, part named BBa_K2492004 is constructed. This part provides an neuron-AWA specific promoter. BBa_K2492001 is a part of channelrodopsin, which serves as a light switch. If any team require an part to manipulate neuron activity through ~470nm excitation light , they can choose this part. BBa_K2492003 is an calcium ions indicator part, it provides the function of monitoring and responding to the concentration of calcium ions. The BBa_K2492002 part codes a kind of fluorescence reporter protein named mcherry. It serves as an reporter, which provides us with direct visualisation of whether plasmid is injected successful or not and where the transgenic neuron is. BBa_K2492000 is another fluorescence protein called GFP. Two reporter proteins are all modified through adding introns, which enables the expression in C.elegans.
